Qatar Airways has ordered two Boeing 747-8 Freighters, and added another four Boeing 777-300ERs to its orderbook. The 747 was already being considered as an airlifter before it was built. As far as the USAF goes, they went with the Lockheed C5. From experience, the C5 is a lot easier to load and unload than a 747. Seems like we would have a lot of broke C5's on the ramp as opposed to civilian 747's. Except for that time someone poked a hole on that one 747 at Ramstein. Oops.

Now wouldn't that make sense. I haven't considered the floor is not removable (without major surgery like the 747 Dreamlifter) and I haven't considered the weight for extra smaller containers needed for smaller clearance (the ones on 747 main deck were very big). Plus A380s' stubby fuselage and large wings.
After all, one problem all planes faces is weight, most planes don't fly with full payload and fuel (which means they fly lot of stuff less fuel or more fuel less stuff), especially on long haul runs.

It's been thought of, but it's a lot harder than you would think. There is no cost effective way for an A380 conversion so Airbus scrapped that idea.
